报告题目 Microscale Mass Spectrometry: Single Cell, Tissue Imaging, and Tumor Spheroid
报告摘要 Mass spectrometry (MS) is a rapidly developing area, and the corresponding techniques have been widely used for sensitive detection and accurate identification of broad ranges of molecules. Sampling and ionization of analyte under ambient conditions are critical for modern MS bioanalysis. We have developed a multifunctional sampling and ionization device, named the Single-probe, that can be coupled to a mass spectrometer for micro-scale bioanalysis in three major areas. (1) Single cell MS. The Single-probe has a small enough tip (<10 μm) that can be directly inserted in live single cells to extract cellular contents, which are ionized for real-time MS analysis. (2) MS imaging. Using the same instrumentation, the Single-probe was used to conduct micro-scale liquid extraction of molecules on tissue sections, and construct the spatial distribution of biomolecules with relative abundances on tissues under ambient conditions. (3) MS depth profiling analysis live tumor spheroids. To better mimic micro-environment in real solid tumors, we conducted 3D culture of tumor spheroids, and treated them using anti-cancer drug. We gradually inserted the Single-probe tip into spheroids to measure the drug penetration.
报告人简介： Zhibo Yang，美国Oklahoma大学化学与生物化学系助理教授。主要从事质谱分析化学研究。在质谱生物分析、活细胞质谱分析、质谱成像、活肿瘤细胞球质谱分析、气相离子化学以及计算化学等多个方面取得一系列研究成果，发表学术论文50余篇，曾获美国质谱学会研究奖等奖项。
